In vivo magnetoFection™

 

The unique solution for in vivo targeted gene delivery


MagnetoFection™ has been developed mainly to solve the problem related to diffusion limited process for in vitro applications and to restricted bioavailability at the target site for in vivo applications. 

In vivo MagnetoFection™ has been designed for in vivo targeted transfection and infection.
This original system combines magnetic nanoparticles and nucleic acid vectors that will be retained after injection at the magnetically targeted site. In this way, targeted delivery minimizes systemic distribution and reduces toxicity.
Furthermore, the magnetic force will enhance the uptake of magnetic nanoparticles by the target tissue, and thus improve the efficiency of transfection/transduction. This allows decreasing the required process time of delivery to few minutes which is crucial for improvement of in vivo nucleic acid delivery.

The main advantages are:

  • High and increased transfection/transduction efficiency

  • Targeted process (magnetically-driven) / Penetration of the vector into tissues

  • Reduction of the systemic dissemination of vectors during injection

  • Reduction of the vector doses (nucleic acid, virus…)

  • Work under non permissive conditions (hypothermia, physiological flow conditions)

In vivo MagnetoFection™ applications  

Nucleic acids and vectors

In vivo MagnetoFection™ has been developed for in vivo targeted transfection of various types of nucleic acids such as DNA, RNA, oligonucleotides or transduction with any viral vectors

OZ Biosciences offers three types of ready-to-use In vivo Magnetofection™ reagents:

  • In vivo PolyMag is a cationic polymer-based magnetic nanoparticles formulation.
    It can be used with plasmid DNA, antisense oligonucleotides, mRNA and siRNA.
  • In vivo DogtorMag is a cationic lipid-based magnetic nanoparticles formulation.
    It associates Dogtor a specific cationic lipid and CombiMag magnetic nanoparticles. This reagent is suitable for plasmid DNA, antisense oligonucleotides, mRNA and siRNA.
  • In vivo ViroMag is an optimized nanoparticles formulation dedicated to viral vectors.
    It is particularly suitable for Lentiviral/Retroviral, Adenoviral and Adeno-Associated Viral (AAV) vectors.  

Several routes of administration

DNA/nanoparticles or virus /nanoparticles complexes can be easily administrated through various injection routes:
- Systemic administration (intravenous, intra-artery)
- Local administration (intratumoral, intracerebroventricular, intraperitoneal, intramuscular, subcutaneous).

All In vivo MagnetoFection™ reagents have been especially designed by our R&D team to meet in vivo grade quality. 

Magnet can be positioned:
- externally for large organs or isolated oragns (kiver, brain, muscle, subcutaneous tumor)
- internally for deep oragns or focalized gene transfer

Success Stories

 Transfection of tumor in cat - Cancer gene therapy

Magnetofection™ has been successfully used in veterinary clinical studies for the treatment of feline fibrosarcoma using immunostimulatory gene therapy. Feline fibrosarcoma is one of the most common feline tumors with a relapse rate of 75% within 6 months upon surgical resection.
Gene therapy was conducted on cats receiving pre-operative intratumoral injection of plasmid encoding granulocyte-macrophage colony-stimulating factor and magnetic nanoparticles. These studies showed no adverse events as well as an increase in animal survival (references 5, 6, 7).

After intratumoral injection of the plasmid-attached nanoparticles (FigA), a magnetic field was applied directly onto the tumor for magnetic guidance of magnetic beads-associated plasmid (FigB).

Transfection of ear artery endothelial cells - Tissue targeting

In this study, Magnetofection™ has been used for local gene delivery in blood vessels. Plasmid DNA encoding luciferase complexed with magnetic nanoparticles were injected into the ear veins of pig.
A permanent magnet was placed upstream the blood flow circulation and near the injection site as shown in the photo hereunder. Results demonstrated that transgene was expressed at the magnet position indicating an efficient magnetic targeting.
