Is Magnetofection suitable for suspension cells and adherent cells?
Yes, Magnetofection is suitable for adherent and suspension cells. We have tailored and optimized a protocol for suspension cells that is detailed in the instruction manual.
Briefly, while PolyMag / DNA complexes or CombiMag / transfection
reagents (or virus) complexes incubate, dilute the cells to be
transfected to 5x10 5 - 1x10 6 / mL in medium (with or without serum or
supplement) and perform one of the following options to sediment the
cells at the bottom of the culture dish in order to promote the contact
with the magnetic nanoparticles.
- Option # 1 : Seed the cells on polylysine-coated plates and use the protocol for adherent cells
- Option # 2 : Briefly centrifuge the cells (2 minutes) in order to pellet them and use the protocol for adherent cells
- Option # 3 : Mix cell suspension with 30µL of CombiMag reagent per 1mL of cell suspension.
a. Incubate for 10-15 minutes
b. Distribute cells to your tissue culture dish placed
upon the magnetic plate (volume of culture medium containing cells
depends on the culture dish size; see suggested transfection volume in
the protocol)
c. Incubate for 15 minutes.
Then add the mixture of transfection complexes to the
cells while keeping the cell culture plate on the magnetic plate.
Continue to incubate for 15 minutes and then proceed as explained in
the instruction manual.
